Chemiluminescence - intensification of involution
In the field of chemiluminescence, the phenomenon of involution is intensifying, and the industry is facing a dilemma. With the continuous development of technology and the intensification of market competition, enterprises and research institutions are facing tremendous pressure and challenges. The causes of chemiluminescence involution are analyzed as follows:
(1) The market side has continuously increased requirements for the degree of equipment automation, and the product price is required to continue to decline, resulting in an increase in enterprise costs, insufficient ability to compress production costs, and a decrease in capital sources, which is a dilemma;
(2) You can't reduce costs quickly, and you can only choose to continue to roll in or gradually go out;
(3) Although the multi-channel single-person luminometer solves the problem of instrument cost, the reagent needs to be refrigerated and stored, and the transportation cost will be greater than the cost of the reagent;

System composition
Two antibody-DNA complexes (antibody 1-DNA1, antibody 2-DNA2), acridine ester (AE)-labeled DNA3 (DNA1, DNA2, and DNA3 single strands each contain 7–9 bases each paired in pairwise pairing), endonuclease, and graphene oxide (GO)
Brief description of the principle
(1) When the sample contains the target antigen, the antibody (Ab) in the two antibody-DNA complexes will recognize the analyte at the same time, and the sandwich structure of the antibody-antigen-antibody makes DNA1 and DNA2 close enough to hybridize the paired bases of DNA1 and 2, and then both DNA1 and DNA2 can hybridize with DNA3 to form an ortho complex.
(2) Once DNA3 forms a double-stranded structure, GO cannot adsorb DNA3, and when hydrogen peroxide is added, the chemiluminescence of AE on the ortho complex cannot be quenched by graphene oxide.
(3) The endonuclease introduced at the same time makes the chemiluminescence signal continuously cyclically amplified.
(4) In the absence of analyte, DNA3 is absorbed on the surface of GO by vulture-vulture stacking, and when hydrogen peroxide is added, the chemiluminescence of acridine ester attached to the end of single-stranded DNA3 is quenched by graphene oxide without affecting the results.
